A murine monoclonal antibody (MAb), specifically recognizing and blocking the Epidermal Growth Factor Receptor, was obtained. To prevent a humoral response against murine antibody when used clinically, a humanized antibody was constructed by combining the complementarity-determining regions of our own murine Abs with human framework and constant regions. In addition, several amino acids in the framework were changed to improve the binding affinity of the antibody and further reduce its potential immunogenicity. The humanized antibody, were docked and recognized that it had affinity of -6.345032e+002 k.cal/mole for human EGFR (1IVO), within two-fold of that of the original murine antibody. These humanized variants are being synthesized and are soon going to be tested on the EGFR by phage typing and ELISA.

Once these tests were successful the humanized antibody will have clinical potency in curing different cancers like lung, ovarian etc
